Description

Description: This assay employs a two-site sandwich ELISA to quantitate FLG2 in samples. An antibody specific for FLG2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FLG2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FLG2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FLG2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The presence of ifapsoriasin (IFPS, fillagrin-2), a novel skin- and mucosaspecific S100 fusion type protein, at the surface of skin and mucosa, particularly where moisture-requiring soil bacteria may grow, reveals its potential as innate defense factor in the protection of body surfaces from infection with soil bacteria.
IFPS may be used in the therapy of infectious diseases of epithelial organs and for infection prophylaxis at interfaces, especially of the skin, the nasopharynx, the eyes as well as the respiratory, gastrointestinal and urogenital tracts. The filaggrin 2 is a new member of the EDC, a cluster of approximately 50 genes involved in cornification. filaggrin 2, a poorly characterized member of the epidermal differentiation complex, as well as three new lipase genes clustered with paralogous genes on chromosome 10q23.31
